12.5 with key off is marginal and indicates a battery hats on its way out the door. Though it could be that it just needs a good charge. In either case, it's not the source of the problem.

12 volts after turning the key on is decent.

Voltage with engine running in the range of 3-4000 rpm should be closer to 14.4 but 14 even is acceptable. Only at idle should it be in the mid 13's.

And yes, it's only running on one cylinder. Check the valves and adjust as necessary before doing anything else. The go after the non running cylinder. Valves should always be done before mucking with the carbs.
